Abstract

The aim of the work was a comparative study of the morphological characteristics of the thyroid gland
(TG), kidney, liver, and blood biochemical indicators (ALT, AST, urea, total and direct bilirubin, creatinine) in
normo- and hypertensive rats with propylthiouracil (PTU) application.
Materials. The experiments were performed on female SHR rats of 6 months of age. Wistar rats were used
as a normotensive control. Hypothyroidism was induced for 39 days using 0.1% PTU added to drinking water.
On different days of the experiment, the rats were killed; blood was taken to determine biochemical indicators
and samples of the thyroid gland, kidney, liver for the preparation of histological slides.
Results. In rats of both groups, a significant (5-fold) increase in thyroid mass was observed in comparison
with intact animals. Histopathologically, the follicular epithelium of TG was characterized by pronounced signs
of hypertrophy and hyperplasia. Fatty vacuolization of hepatocytes and focal necrosis were observed in the liver.
The appearance of venous hyperemia in the TG and the liver was noted with the intake of PTU. No obvious
pathological changes were detected in the kidneys. SHR rats were found to be more sensitive to the thyrostatic
and hepatotoxic effects of PTU than Wistar rats. This was evidenced by a significant increase in the height of
the follicular epithelium and the number of pathological follicles with papillary structures, an increase in the
levels of ALT, AST, and an increase in the average area of the central vein of the liver in hypertensive rats.
Thus, PTU application induces characteristic morphological changes in the TG in SHR rats, which allows the
use of rats of this line as model animals when studying thyroid dysfunction against hypertension.
