Influence of extract of peach ordinary (persica vulgaris) leaves on the state of the hypothalamic-pituitary-adrenal system of rats in conditions of chronic immobilization stress

Abstract

Study of the influence of thick extract of peach ordinary (Persica vulgaris) leaves (TEPL) (patent 103001, Ukraine), which was obtained at the Department of Chemistry of Natural Compounds of National University of Pharmacy on the state of the hypothalamic-pituitary-adrenal system (HPAS) of rats in conditions of chronic immobilization stress. The model of chronic immobilization stress was reproduced for 18 days by daily four hour immobilization of rats in tight boxe. Investigated TEPL was administered intragastrically in a conditionally ef­fective dose of 100 mg/kg. Reference drug the syrup «Immuno-Ton» was used in a dose of 3 ml/kg. The state of the HPAS was determined by the coefficients of the mass of the adrenal glands and thymus, the degree of ulcer­ation in the stomach and the results of histological study of the structure of the adrenal glands. Established the protective effect of TEPL on the state HPAS. This is confirmed by a decrease in the degree of ulcerative damage to the stomach, a decrease in thymus involution and adrenal glands hypertrophy. TEPL reduced morphological changes in the adrenal tissues of rats compared with untreated animals due to inhibition of increased activity of adrenocorticocytes of the cortex, neuroendocrinocytes of the medulla, as well as a decrease in morphometric parameters: the width of the beam zone and the diameter of the nuclei of the adrenocorticocytes of the beam zone. The protective effect TEPL was at the level of the «Immuno-Tone» comparator drug. The obtained results substantiate the expediency of further research on the pharmacodynamics and mechanisms of action of TEPL for the development on its basis of a new drug of stress-protective action.
